Rapid Determination of Lignans in Health Products of Schisandra chinensis by Ultra-High Performance Liquid Chromatography-Quadrupole/Time-of-Flight Mass Spectrometry
Objective:To establish an ultra-high performance liquid chromatography-quadrupole/time-of-flight mass spectrometry method for simultaneous determination of schisandrin,schisandrin B and schisandrin A in health food containing Schisandra chinensis.Method:Samples were extracted with 70%acetonitrile solution,salted out and stratified,and the combined acetonitrile layer was purified with 80 mg C18 solid phase adsorbent.Under optimized instrument conditions,ESI ionization source was used for scanning in positive ion mode,and APCI ionization source was used for correction.Result:The linear relationship between the three lignans was good in the range of 50~2 000 μg·L-1,the correlation coefficients were all≥0.999 0,the average recoveries were 87.0%~104.1%,the relative standard deviations were 2.98%~6.56%,the limits of detection were 1.0 mg·kg-1,and the limits of quantification were 2.5 mg·kg-1.Conclusion:The method has simple pretreatment steps,less consumables,high recovery,sensitivity,selectivity and reproducibility,and can be used for rapid determination of lignan content in health food containing Schisandra chinensis.
