Screening and Characterization of Uric Acid-Reducing Lactic Acid Bacteria
Objective:To screen lactic acid bacteria with the potential to reduce uric acid from natural fermentation samples.Method:The nucleoside degradation rate and xanthine oxidase(XOD)inhibition of isolated strains were measured and screened.A high uric acid zebrafish model was established to evaluate the uric acid lowering effect of the strains,and the strain combination ratio was optimized.Result:The isolated and screened Lactobacillus helveticus GQ1501 can achieve 100%degradation of inosine and guanosine;the inhibition rates of Lacticaseibacillus paracasei R2 and Lactiplantibacillus plantarum D1 on XOD were 44.05%and 37.61%,respectively;the evaluation of the high uric acid zebrafish model showed that strains GQ1501,R2,and D1 exhibited good uric acid lowering effects in zebrafish.The optimal combination ratio was R2∶GQ1501∶D1=2∶1∶1,and the uric acid degradation rate reached 53.07%at a concentration of 0.5 g·L-1;compound strains can impart various flavor compounds to fermented milk,among which 2-nonanone has the highest content of 86.27 mg·kg-1.Conclusion:The strains and their combinations isolated and screened in this study have the potential to develop uric acid reducing fermented milk.
