摘要
本研究系统分析了不同培养条件下甲壳素酶活性及蛋白质含量的变化,特别关注了葡萄糖添加量、蛋白质添加量、甲壳素添加量、接种量和培养条件对微生物生长、酶活力及蛋白质合成的影响.结果显示,培养基中葡萄糖添加量为4 g·L-1、蛋白胨添加量为16 g·L-1、甲壳素添加量为10 g·L-1、青霉接种量为1.0×105个/L,pH值5.5、振摇转速180 rmin-1、自然通气、温度45 ℃的条件下,培养4d,所产甲壳素酶的活力最高,达到57.6U·mL-1,蛋白含量为620.7mg·mL-1.综合来看,不同条件的优化组合能够有效提升海洋青霉的酶产量,为工业化应用提供了科学依据.
Abstract
This study systematically analysed the changes in chitinase activity and protein content under different culture conditions,focusing on the effects of glucose,peptone,chitin,inoculation amount and culture conditions on microbial growth,enzyme activity and protein synthesis.The results show that,under the conditions of 4 g·L-1 glucose,16 g·L-1 peptone,10 g·L-1 chitin,1.0×105 per liter of Penicillium oxalicum,pH 5.5,180 r·min-1 shaking speed,natural ventilation and 45 ℃ for 4 days,the enzyme of chitinase had the highest enzyme activity of 57.6 U·mL-1,corresponding to a protein content of 620.7 U·mL-1.The optimised combination of conditions improves the enzyme yield of Penicillium oxalicum,providing a basis for industrial application.
