Determination of Diclazuril,Toltrazuri and Its Metabolites in Foods of Animal Origin by HPLC-MS/MS

目的:建立一种适用于测定动物源食品中地克珠利、妥曲珠利及其代谢物残留量的方法.方法:样品用乙腈提取后,经PSA净化,液相色谱-串联质谱检测.结果:在1.0~100.0 μg·L-1线性范围内,4种待测物相关系数(r2)为0.997 363~0.999 085,水产品、畜禽肌肉、禽蛋类样品检出限均为2.5 μg·kg-1,内脏样品检出限均为5.0 μg·kg-1.当地克珠利、妥曲珠利及其代谢物添加浓度为12.5~250.0 μg kg-1时,在鱼肉样品中回收率为70.01%~88.83%,在鸡肉样品中回收率为70.60%~89.68%,在鸡蛋样品中回收率为74.83%~88.72%;当添加浓度为25.0~500.0 μg·kg-1 时,猪肝样品回收率为70.84%~85.34%,相对标准偏差(n=6)为0.80%~5.35%.结论:本方法操作简单、灵敏度高,适用于测定动物源食品中地克珠利、妥曲珠利及其代谢物的残留量.
Objective:To establish a method for the determination of diclazuril,toltrazuril and their metabolites in animal-derived foods.Method:The samples were extracted with acetonitrile,cleaned up by PSA and detected by liquid chromatography-tandem mass spectrometry.Result:The correlation coefficients(r2)of the four analytes were 0.997 363~0.999 085 in the linear range of 1.0~100.0 μg·L-1.The detection limits of aquatic products,livestock and poultry muscle,and poultry eggs were all 2.5 μg·kg-1,and the detection limits of viscera samples were all 5.0 μg·kg-1.When the concentrations of diclazuril,toltrazuril and their metabolites were added at 12.5~250.0 μg·kg-1,the recoveries in fish samples were 70.01%~88.83%,in chicken samples were 70.60%~89.68%,and in egg samples were 74.83%~88.72%.When the addition concentration was 25.0~500.0 μg·kg-1,the recovery rate of pig liver samples was 70.84%~85.34%,and the relative standard deviation(n=6)was 0.80%~5.35%.Conclusion:This method is simple to operate and has high sensitivity.It is suitable for the determination of diclazuril,toltrazuril and their metabolites in animal-derived foods.
