目的:研究八珍粉对脾胃的改善作用.方法:动态随机选取3dpf(受精后3d)斑马鱼,以水溶方式给予三硝基苯磺酸构建胃肠黏膜损伤模型,建模成功后以水溶方式分别给予八珍粉和泼尼松,用阿尔新蓝染色,通过分析斑马鱼肠道杯状细胞数量评价八珍粉对胃肠黏膜损伤的修复功效;动态随机选取5 dpf(受精后5d)斑马鱼,以水溶方式给予硫酸铝建立便秘模型,建模成功后以水溶方式给予八珍粉和多潘立酮,通过分析斑马鱼肠道标记强度评价八珍粉对便秘的改善功效.结果:1 000、2 000 μg·mL-1八珍粉对斑马鱼胃肠黏膜损伤具有显著的改善作用,且呈现剂量相关性;500、1 000、2 000 μg mL-1八珍粉对斑马鱼便秘具有显著的改善作用,且现呈剂量相关性.结论:八珍粉对斑马鱼脾胃有一定的改善作用.
Objective:To investigate the effects of eight-treasure powder on the improvement of spleen and stomach functions.Methods:3 dpf(3 days post-fertilization)zebrafish are randomly selected in a dynamic manner to construct a gastrointestinal mucosal injury model by giving tritrobenzene sulfonate in a water-soluble manner.After successful modeling,eight-treasure powder and prednisolone are given in water solution,respectively.The repair effect of eight-treasure powder on gastrointestinal mucosal injury is evaluated by analyzing the number of intestinal cup-cells in zebrafish stained with Alcian blue.5 dpf(5 days post-fertilization)zebrafish are randomly selected dynamically and given aluminum sulfate in water solution to establish a constipation model.After successful modeling,eight-treasure powder and domperidone are given in the way of water solution,and the improvement effect of eight-treasure powder on constipation is evaluated by analyzing the intestinal fluorescence intensity of the zebrafish.Result:1 000,2 000 μg·mL-1 eight-treasure powder significantly improve the gastrointestinal mucosal injury of zebrafish,and show dose correlation.500,1 000,2 000 μg·mL-1 eight-treasure powder has a significant effect on zebrafish constipation,and it is dose-dependent.Conclusion:Eight-treasure powder can improve the spleen and stomach of zebrafish to some extent.
