高效液相色谱法测定发酵饲料中乳酸的含量

建立了一种能准确测定发酵饲料中乳酸含量的液相色谱分析方法.以C18柱为分析柱,采用紫外检测器,10 min内能够很好地分离出乳酸.流动相为磷酸二氢钾溶液,检测波长为210 nm,外标法定量测定.实验结果表明,该方法在1~300 mg/L的浓度范围内,线性关系良好,R2>0.999.检出限为30 mg/kg,定量限为50 mg/kg,加标回收率为86.6%~98.0%.该方法灵敏度和准确度高、重复性好,可适用于发酵饲料中乳酸的测定.
Determination of lactic acid in fermented feed by high performance liquid chromatography
A liquid chromatography method was developed for the accurate determination of lactic acid content in fermented feed.Using C18 column and UV detector,lactic acid could be well separated in 10 minutes.The mobile phase was potassium dihydrogen phosphate solution,the detection wavelength was 210 nm.The external standard method was used for quantitative determination.The method had a good linearity in the concentration range of 1 mg/L to 300 mg/L,R2>0.999.The limit of de-tection was 30 mg/kg,the limit of quantitation was 50 mg/kg and the recovery rate of standard addition were 86.6%-98.0%.This method had high sensitivity,accuracy and good repeatability,and was suitable for the determination of lactic acid in fer-mented feed.
