响应面法优化假单胞菌koreensis产聚-β-羟基丁酸酯的发酵工艺参数

Optimization of Fermentation Parameters for poly-β-hydroxybutyrate Pro-duction of Pseudomonas sp. koreensis by Response Surface Methodology

聚-β-羟基丁酸酯(PHB)作为一种完全可降解生物塑料,有望替代传统塑料应用于食品包装、农业和医药等领域,但高昂的生产成本制约着PHB更广泛的应用。本文在以前培养基配方优化结果的基础上,对发酵工艺参数进行响应面优化,确定最佳发酵工艺参数,进一步提高PHB产量。首先,通过单因素试验,确定了转速、初始pH值、接种量和温度的水平范围;然后,以单因素试验结果为基础,采用Design Expert8.0软件设计Box-Behnken四因素三水平、5个中心点、共29组的响应面试验,得到拟合回归方程;最后,通过软件分析确定了假单胞菌koreensis发酵产PHB的最佳工艺参数。结果表明:转速170.91r/min、初始pH值7.64、接种量6.21%和温度32.15℃, PHB产量达4.42g/L,为优化前的1.38倍,效果显著。
As a completely biodegradable plastic, poly-β-hydroxybutyrate ( PHB) is expected to replace traditional plastics used in food packaging, agriculture, medicine, etc., but high cost of production restricts the wider applications of PHB. Based on previous results on media formulations, optimum fermentation parameters were determined by response surface method-ology, and yield of PHB was promoted. First of all, level range of rotation speed, initial pH, inoculation amount and tempera-ture were confirmed; and then, fitted regression equation was gotten based on results of single factor experiments by using Design Expert8.0 to design Box-Behnken response surface experimental; finally, optimum fermentation parameters were determined by software analysis. Results showed that rotation speed 170.91r/min, initial pH 7.64, inoculation amount 6.21%, and temperature 32.15℃, PHB production reached 4.42g/L. Effect was significant that production of PHB was 1.38 times as before.
