Research on the physiological changes in plants to adapt to stressful environments and their ability to adapt to stressful environments has been a hot issue in recent years.Osmoregulation is the main physiological mechanism for plants to adapt to stressful environments,among which the determination and study of the osmotic potential of plant leaves is the key to determine the osmoregulatory capacity of plants.In this study,the osmotic potential of plant leaves was determined by the occurrence of the phenomenon of plasmatic wall separation after incubation with a gradient concentration of sucrose solution using arabidopsis,lemongrass and larch as the subjects.The isotonic concentrations of Arabidopsis thaliana,Caragana microphylla and Larix gmelinii were 0.375,0.575 mol/L and 0.825 mol/L(sucrose solution)respectively.The calculated cellular osmotic potential was-921.3,-1 412.66,and-2 026.86 kPa for Arabidopsis,lemongrass,and Larix gmelinii,respectively.The determination of the osmotic potentials of the leaves of Arabidopsis thaliana,Caragana korshinskii microphylla and Larix gmelinii was useful for improving the efficiency of plant tissue culture and plant survival.At the same time,it provides theoretical basis for the study of osmoregulation mechanism of plants and the selection and breeding of excellent varieties.
