Effects of PsWOX11 on Lateral Root Growth and Development of Populus simonii
[Objective]The effects of PsWOX11 gene on the growth and development of lateral root in Pop-ulus simonii were analyzed to provide reference for exploring the molecular mechanism of root growth and development regulated by WOX11 gene in woody plants.[Method]Bioinformatics methods were used to analyze gene and protein structures,and conserved motifs.The tissue-specific expression patterns were analyzed by qRT-PCR.Overexpression(35S∷PsWOX11,OE)and dominant repression(35S∷PsWOX11-SRDX,DR)transgenic PsWOX11 poplar plants were generated and used to analyze the effects of PsWOX11 gene on the growth and development of lateral root.[Result]The PsWOX11 gene,with 768 bp in length and encoding a 255 amino acid protein,was cloned from Populus simonii.The expression level of PsWOX11 gene was relatively high in the main roots and lateral roots of poplar.Transgenic poplar lines overexpressing PsWOX11(OE1-OE7)and PsWOX11 dominant repression plants(SRDX1-SRDX8)were generated,and the phenotype and microscopic morphology of different genotypes of poplars showed that the lateral root length and diameter of transgenic poplars overexpressing PsWOX11 were significantly higher than those of non-transgenic 84K poplar plants,while opposite phenotypes were observed for PsWOX11 dominant repression plants.[Conclusion]PsWOX11 gene is involved in regulating the growth and development of lateral roots in poplar,which increases the length and diameter of lateral roots.This study provides a reference for further understanding that WOX11 gene is involved in regulating root growth of poplar.
