Determination of hydroxy safflower yellow A in natural dyes by high performance liquid chromatography-tandem mass spectrometry
In order to explore the identification method of hydroxyl safflower yellow A in natural dyes, which is distinguished from chemical synthetic dyes,a method for the quantitative analysis of hydroxyl safflower yellow A in natural dyes was developed by high performance liquid chromatography-tandem mass spectrometry. The samples were qualitatively and quantitatively analyzed in the multi reaction monitoring mode(MRM),25%methanol aqueous solution was used for extraction, the flow rate of mobile phase was 0.25 mL/min,the sample volume was 5 μL,the column temperature was 40℃,and methanol-10 mmol/L ammonium acetate aqueous solution was used as mobile phase, then separated by Perkin Elmer column C18(2.7 μm,2.1 mm×100 mm)The results show that the linear relationship is good when the mass concentration is 0.02-2.00 mg/L,and the fitting degree is higher than 0.999 0.The limit of detection was 0.125 μg/L,and the limit of quantitation was 0.42 μg/L,and the recovery was between 80%and 95%at different concentrations; This method can provide an efficient and sensitive detection scheme for the determination of hydroxyl safflower yellow A,and also provide a reference for the product quality determination of natural dye-dyed textiles.
