摘要

目的 探讨circRNA_0008720对乳腺癌细胞增殖、侵袭的影响及其可能的作用机制.方法 采用实时荧光定量PCR(qRT-PCR)技术检测乳腺癌组织中circRNA_0008720、miR-138-5p、SOX4的表达水平,并通过荧光素酶报告基因实验明确三者之间的靶向关系.通过CCK-8、Transwell和Western blot检测乳腺癌细胞的上皮细胞-间充质转化(EMT)程度以及增殖、侵袭能力.结果 circRNA_0008720、SOX4在乳腺癌组织和乳腺癌细胞系中均表达上调,miR-138-5p表达降低(均P<0.05).敲低circRNA_0008720或上调miR-138-5p的表达可以抑制乳腺癌细胞的EMT进程和增殖、侵袭活性(均P<0.05),而抑制miR-138-5p或过表达SOX4能够部分逆转这种作用.结论 circRNA_0008720可通过海绵化miR-138-5p上调SOX4表达,促进乳腺癌细胞的EMT进程和增殖、侵袭活性.

Abstract

Objective To investigate the effect of circRNA_0008720 on the proliferation and invasion of breast cancer cells and its possible mechanism.Methods Real-time quantitative fluorescent PCR(qRT-PCR)was used to detect the expression of circRNA_0008720,miR-138-5p and SOX4 in breast cancer tissues,and the targeting relati-onship between them were determined by luciferase reporter gene assay.The EMT degree,proliferation and invasion ability of breast cancer cells were detected by CCK-8,Transwell and Western blot.Results circRNA_0008720 and SOX4 were up-regulated in breast cancer tissues and breast cancer cell lines,while miR-138-5p expression was de-creased.Knockdown the expression of circRNA_0008720 or up-regulate miR-138-5p could inhibit EMT process,proliferation and invasion activity of breast cancer cells;while inhibition of miR-138-5p or overexpression of SOX4 can partially reverse this effect.Conclusions circRNA_0008720 can up-regulate SOX4 expression through spon-ging miR-138-5p to promote EMT process,proliferation and invasion activity of breast cancer cells.
