黄连素液通过影响NF-κB信号通路修复小鼠细菌感染皮肤创面的作用机制研究

目的 探讨黄连素液通过影响核因子(NF)-κB信号通路修复小鼠细菌感染皮肤创面的作用机制.方法 选取60只SPF级9周龄雄性C57BL/6J小鼠构建耐MRSA皮肤创面感染模型,将造模成功的50只小鼠随机分为模型组、阳性对照组以及黄连素液低、中、高剂量组,每组10只.分别于给药第3、7、14天计算各组小鼠创面愈合率并测定创面菌落数.给药结束后观察各组小鼠创面组织病理变化;检测各组小鼠创面组织白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)、基质金属蛋白酶-2(MMP-2)及血管内皮生长因子(VEGF)的表达.检测Toll样受体2(TLR2)、转化生长因子-β活化激酶结合蛋白1(TAB1)、NF-κB p65的mRNA及蛋白表达.结果 随黄连素剂量增加,模型组创面愈合率逐渐提升(均P<0.05);菌落数逐渐减少(P<0.05).模型组淋巴细胞、中性粒细胞丰富,肉芽增生显著;低剂量组肉芽轻度增生,炎细胞轻度浸润;中剂量组无肉芽增生,炎细胞少量;高剂量组仅见少量中性粒细胞.随黄连素剂量增加,IL-6、TNF-α、MMP-2水平下降,VEGF水平上升,TLR2、TAB1及NF-κB p65的mRNA及蛋白表达逐渐降低(均P<0.05).结论 黄连素液可能通过抑制TLR2/NF-κB信号通路激活,减轻小鼠MRSA感染皮肤创面组织炎症反应,进而促进创面愈合.
Objective To explore the mechanism of Berberine liquid in repairing bacterial infected skin wounds in mice by affecting the nuclear factor(NF)-κB signaling pathway.Methods Sixty nine-week-old male SPF C57BL/6J mice were selected to construct a methicillin-resistant staphylococcus aureus(MRSA)model.Fifty mice were ran-domly divided into model group,positive control group and Berberine liquid low(1.25%),medium(2.5%)and high dose groups(5%),with ten mice in each group.The healing rate and the number of colonies were calculated on the 3rd,7th and 14th day of administration,respectively.The pathological changes of wound tissue were observed after administration.The levels of interleukin-6(IL-6),tumor necrosis factor α(TNF-α),matrix metalloproteinase-2(MMP-2)and vascular endothelial growth factor(VEGF)were detected by enzyme-linked immunosorbent assay(ELISA).The mRNA expressions of toll-like receptor 2(TLR2),transforming growth factor-β-activated kinase-bin-ding protein 1(TAB 1)and NF-κB p65 were detected by RT-qPCR.The protein expressions of TLR2,TAB 1 and NF-κB p65 were detected by Western blot.Results With the duration of administration,the wound healing rate of mice in model groups gradually increased(P<0.05),the number of clonies gradually decreased(P<0.05).HE staining showed that the model group had significant granulation and more inflammatory cells.The low dose grop had a mild granulation and inflammatory cells;The medium dose group had no granulomatous tissue hyperplasia and slight in-flammatory cells.A few neutrophils were found in the high dose group.With the increase of administration,the ex-pression of IL-6,TNF-α and MMP-2 were decreased,and VEGF level was increased(all P<0.05);the expressions of TLR2,TAB1,NF-κB p65 mRNA and protein were decreased(all P<0.05).Conclusions Berberine liquid may attenuate the tis-sue inflammatory response of MRSA-infected skin wounds in mice by inhibiting the activation of the TLR2/NF-κB signa-ling pathway,and promote wound healing.
