山药皮提取物中功效成分的分离纯化研究

采用重结晶和柱分离法对山药皮提取物中功效成分尿囊素和皂苷进行同步分离试验,研究结果表明,选用AB-8型大孔树脂为皂苷的吸附树脂,吸附流速2mL/min;吸附完成后用70%的乙醇进行洗脱,其洗脱率可达90%;收集洗脱液并对其进行真空浓缩后,再进行真空干燥,得淡黄色的皂苷产品,经法定机构检测,其纯度为71.5%,得率为0.17%;将含尿囊素的浓缩液进行结晶,温度为0~4℃,时间为24 h.所得晶体再以水作为溶剂,在料液比为1∶3的情况下重结晶2次,干燥后得到纯度为85.8%的尿囊素产品,得率为0.25%.
Study on Separation and Purification of Functional Components in Chinese Yam Peel Extract
Using the recrystallization and column separation method for effect of Chinese yam peel extract components of allantoin and saponin sync separation.The results show that the selected AB-8 macroporous resin adsorption resin saponin,adsorption velocity is 2 mL/min;adsorption finished with 70% ethanol elution,eluting rate in more than 90%;washing liquid to collect and vacuum concentrating on its,then vacuum drying,light yellow saponin product,by legal institutions detection,its purity 71.5%,the yield is 0.17%;concentrate containing allantoin to crystallization,temperature is 0~4 ℃,time 24 h.The crystals with water as solvent,ratio of material to liquid is 1 ∶ 3 in case 2 times recrystallization,drying to obtain allantoin product purity of 85.8%,the yield is 0.25%.
