Effects of miR-519b-3p on the apoptosis,migration and invasion of cervical cancer cells by regulating IL-6/STAT3 pathway
Aim To investigate the effect of miR-519b-3p on apoptosis,migration,and invasion of cervical cancer cells(Si-Ha)through the regulation of IL-6/signal transduction and transcription activating factor 3(STAT3)pathway.Methods qRT-PCR was used to detect the expression of miR-519b-3p in cervical cancer tissue.SiHa cells were divided into miR-NC group,miR-519b-3p group,miR-NC+RhIL-6 group,and miR-519b-3p+RhIL-6 group.Flow cytometry was used to evaluate the apoptosis rate of SiHa cells,while Transwell assay was used to detect cell migration and invasion.Western blotting was used to detect the levels of B-cell lymphomatoma-2 gene(Bcl-2),E-cadherin,N-cadherin,Bel related x protein(Bax),STAT3,and p-STAT3 protein.Re-sults Compared with adjacent tissues,the expression of miR-519b-3p in cervical cancer tissue was downregulated(P<0.05).The apoptosis rate of SiHa cells,Bax and E-cadherin protein levels were higher in the miR-519b-3p group than in the miR-NC group,lower in the miR-NC+RhIL-6 group than in the miR-NC group,and lower in the miR-519b-3p+RhIL-6 group than in the miR-519b-3p group(P<0.05).The migration number,invasion number,Bcl-2,N-cadherin,p-STAT3 protein levels of SiHa cells were lower in the miR-519b-3p group than in the miR-NC group,higher in the miR-NC+RhIL-6 group than in the miR-NC group,and higher in the miR-519b-3p+RhIL-6 group than in the miR-519b-3p group(P<0.05).Conclusion miR-519b-3p inhibits the migration and in-vasion of cervical cancer cells and increases cell apoptosis by inhibiting the IL-6/STAT3 pathway.
