circ_0000502 regulates the proliferation and apoptosis of papillary thyroid cancer cell line TPC-1 by targeting miR-1231
Aim To investigate the effects of circ_0000502 on the proliferation,migration,invasion and apoptosis of TPC-1 cells in papillary thyroid cancer.Methods 26 cases of papillary thyroid cancer tissues and corresponding paracancer tissues were selected.TPC-1 cells were divided into NC group,si-NC group,si-circ_0000502 group,miR-NC group,miR-1231 group,si-circ_0000502+anti-miR-NC group and si-circ_0000502+anti-miR-1231 group.qRT-PCR was used to detect the expression levels of circ_0000502 and miR-1231.MTT was used to detect cell viability.Cell migration and invasion were detected by Transwell system.apoptosis was detected by flow cytometry.The expressions of proliferating cell nuclear antigen(PCNA),matrix metalloproteinase(MMP)-2,MMP-9,B cell lymphoblastoma-2(Bcl-2)and Bcl-2-associated X protein(Bax)were detected by Western blotting.The targeting relationship between circ_0000502 and miR-1231 was detected by dual-luciferase reporter gene assay.Results The expression levels of circ_0000502 was high and the expression of miR-1231 was low in papillary thyroid cancer tissues and cells.Si-lencing circle_0000502 or overexpression of miR-1231 reduced the proliferative vitality,migration,and invasion number of TPC-1 cells,inhibited the expression of PCNA,MMP-2,MMP-9,and Bcl-2 proteins,promoted cell apoptosis,and Bax protein expression(P<0.05).Downregulation of miR-1231 expression reversed the effects of silencing circ_0000502 on TPC-1 cell proliferation,mi-gration,invasion and apoptosis(P<0.05).Conclusion Silencing circ_0000502 regulates the proliferation,migration,invasion and apoptosis of TPC-1 cells by targeting up-regulation of miR-1231.
