The effect and mechanism of ginkgo biloba extract on renal interstitial fibrosis in rats with u-nilateral ureteral obstruction
Aim To explore the effect of ginkgo biloba extract(GBE)on renal interstitial fibrosis(RIF)in rats with unilat-eral ureteral obstruction(UUO)and mechanisms.Methods 20 SD rats were randomly selected as Sham group.54 SD rats were treated with UUO and randomly divided into model group,GBE group and irbesartan group,with 18 rats in each group.The pathological changes of renal tissue were observed,and the levels of serum creatinine(SCr),urea nitrogen(BUN),superoxide dis-mutase(SOD),malondialdehyde(MDA),reactive oxygen species(ROS),and the expression of fibronectin(FN),α-smooth muscle actin(α-SMA),collagen I(Col-I)and hepatocyte growth factor(HGF)in renal tissue,were compared in each group.Results Compared with the Sham group,the levels of SCr,BUN,24 h urinary protein,MDA,ROS and the expressions of FN,α-SMA,Col-I and TGF-β1 were significantly increased in the other group,while the levels of SOD and HGF were significantly decreased(P<0.05).Compared with the model group,the levels of SCr,BUN,24 h urinary protein,MDA,ROS and the expressions of FN,α-SMA,Col-I and TGF-β1 in GBE group and irbesartan group were significantly decreased,while the levels of SOD and HGF were significantly increased(P<0.05).The levels of SCr,BUN,24 h urine protein,MDA,and ROS in the GBE group were lower than those in the irbesartan group,while SOD was higher in the irbesartan group(P<0.05).Conclusion GBE can inhibit the occur-rence and progress of RIF and protect renal function in UUO rats,and its mechanism may be related to correcting the imbalance of hep-atocyte growth factor(HGF)and transforming growth factor-1(TGF-β1).
