Effect of DIAPH3 on EMT and tight junction protein expression in gastric mucosal epithelial GES-1 cells induced by cagA+Hp
Aim To investigate the effect of diaphanous-related formin 3(DIAPH3)on epithelial to mesenchymal transforma-tion(EMT)and tight junction protein expression in gastric epithelial GES-1 cells induced by cytotoxin-related protein(cagA+)and Heli-cobacter pylori(Hp).Methods H.pylori J99 strain(cagA+Hp)was co-cultured with GES-1 cell line.The cell proliferation,in-vasion and apoptosis were observed,and the changes of interstitial transition(EMT)related proteins and tight junction proteins were de-tected.Results Compared with the control group,the OD570 value and the number of invasive cells in the model,DIAPH3-NC,and DIAPH3 groups increased at different times after intervention,and apoptosis rate decreased.The relative expressions of N-cadherin and Vimentin increased.The relative expression of E-cadherin and tight junction proteins(E-Cadherin,claudin-1,ZO-1)decreased(P<0.05);Compared with the model group and the DIAPH3-NC group,the OD570 value,the number of invasive cells,apoptosis rate,and the relative expression levels of N-cadherin and Vimentin decreased in the DIAPH3 group.The relative expression of E-cadherin,occlu-din,claudin-1 and ZO-1 increased(P<0.05).Conclusion Inhibition of NIAPH3 expression can inhibit EMT in gastric epithelium GES-1 cells,prevent cell invasion and metastasis,increase apoptosis rate,and up-regulate the expression of tight junction protein.
