Relationship between polymorphisms of TLR7 gene and susceptibility and severity of hand-foot-mouth disease in children
Aim To explore the relationship between polymorphisms of Toll-like receptor 7(TLR7)gene and susceptibility and severity of hand-foot-mouth disease(HFMD)in children.Methods A total of 304 children with enterovirus 71(EV71)in-fection and HFMD were enrolled as HFMD group.According to disease severity,they were divided into mild group and severe group.A total of 300 healthy children during the same period were enrolled as control group.The genotypes and gene frequencies of TLR7 gene at rs3853839,rs179016 and rs179009 loci in different groups were compared.Whether gene polymorphisms could be risk fac-tors of HFMD was determined by Logistic regression analysis.The mRNA level of TLR7 in peripheral blood mononuclear cells in chil-dren with or without C gene was compared.Results The frequencies of girls'and boys'C genotype in TLR7 gene at rs3853839 and rs179016 loci in HFMD group and severe group were higher than those in control group and mild group(P<0.05).C genotype of TLR7 gene at rs3853839 and rs179016 loci was a risk genotype of HFMD and serious illness(P<0.05).The expression level of TLR7 mRNA in mild and severe HFMD children with C genotype at rs3853839 and rs179016 loci was lower than that in children with-out C genotype(P<0.05).Conclusion C genotype of TLR7 gene at rs3853839 and rs179016 loci is an independent risk factor for HFMD in children,which is correlated with the disease severity.
