Prediction of β-sitosterol as medicinal component in traditional Chinese medicine and its effects on the proliferation and migration of irradiated cervical cancer cells
Aim To predict the targeted key medicinal components of traditional Chinese medicine for differentially expressed genes in cervical cancer,and analyze their effects on the proliferation and migration of irradiated cervical cancer cells.Methods Differentially-targeted traditional Chinese medicine as well as its medicinal components and corresponding targets for cervical cancer were searched through Coremine Medical,TCMSP,Uniprot,and Cytoscape databases,and core targets and key medic-inal components were screened.The protein interactions between target genes corresponding to key medicinal ingredients and differen-tially expressed genes in cervical cancer were analyzed through the STRING database.The effects of key medicinal ingredients on the proliferation and migration,deoxyribonucleic acid(DNA)breakage level of cervical cancer HeLa,Siha cells were detected by using MTT,scratch tests and Western blotting.Results 9 effectively targeted traditional Chinese medicines have been predicted to dif-ferentially expressed genes in cervical cancer.96 medicinal ingredients and 252 target genes were obtained,among which the key medicinal ingredient was β-sitosterol and 10 corresponding core target genes were screened out.There was the protein interaction be-tween the core targeted-genes of β-sitosterol and the differential-genes of cervical cancer.β-sitosterol inhibited the proliferation and migration of cervical cancer cells before and after irradiation,promoted DNA fragmentation levels in HeLa cells of cervical cancer and Siha cells.Conclusion The key medicinal component of targeted traditional Chinese medicine for predicting differentially ex-pressed genes in cervical cancer was β-sitosterol,which could inhibit the proliferation and migration of irradiated cervical cancer cells.
