Effects of polystyrene nanoplastics combined with uranium exposure on growth,development and motor behavior of zebrafish
Aim To explore the effects of combined exposure of polystyrene nanoplastics(PS-NPs)and uranium(U)on the growth,development and motor behavior of zebrafish.Methods Healthy embryos 2 hours post-fertilization were randomly di-vided into blank control group(Con group),U alone exposure group(U group),PS-NPs alone exposure group(PS-NPs group),and PS-NPs and U combined exposure group(PS-NPs+U group).Embryo survival rate,hatching rate and malformation rate of each group were recorded.Body length,head area and yolk sac area of larva were measured by Image J.The zebrafish behavior monitor was used to measure the behavioral changes of larva during the day and night.The expression levels of growth and development-relat-ed genes were detected by qRT-PCR.Results Compared with Con group,the embryo survival rate and hatching rate of U group were significantly decreased(P<0.05),and the malformation rate of larva were increased,accompanying with severe dysplasia,re-flected as the decrease of body length,head area and increase of yolk sac area(P<0.05).The larva in U group showed high activity in day and night,as well as decreased expressions of GH,IGF and BMP(P<0.05).The survival,hatching and development of em-bryos and larvae in PS-NPs group showed the same trend as those in U group,but the movement ability was decreased day and night,and the expression of IGF was decreased(P<0.05).The malformation rate of larva in PS-NPs+U group was increased(P<0.05),the motor behavior was reflected as the decrease of motor ability,and the expression of BMP was increased(P<0.05).The combina-tion of PS-NPs and U interfered with the dysplasiacaused by U exposure alone,leading to the decline in motor ability.The expression of GH,IGF,and BMP was upregulated compared with the U group(P<0.05).Conclusion The combined exposure of PS-NPs and U can cause dysplasia and decreased motor ability of zebrafish.PS-NPs interferes with the toxic effect of U,and 200 μg/L PS-NPs and 150 μg/L U showed antagonistic toxicity.
