Curcumin alleviates 6-hydroxydopamine-induced oxidative damage in Parkinson's cell model by regulating miR-221
Aim To study the effect of curcumin on 6-hydroxy dopamine(6-OHDA)-induced oxidative damage in Parkinson(PD)cell model by regulating miR-221.Methods PC12 cells were divided into normal control group,PD group,and curcumin group.After transfection,PD cells were further divided into PD+miR-NC group and PD+miR-221 group,PD+anti-miR-NC+curcumin group and PD+anti-miR-221+curcumin group.Apoptosis was detected by flow cytometry;the expression of Caspase-3 pro-tein was detected by Western blotting;the levels of reactive oxygen species(ROS)were determined by colorimetry,and the activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)were determined by spectrophotometer;qRT-PCR was used to detect the expression level of miR-221 in cells.Results Compared with normal control group,apoptosis rate,expression of Caspase-3 protein,level of ROS and content of MDA were increased in PD group,while activity of SOD and level of miR-221 were de-creased(P<0.05).Compared with PD group,the apoptosis rate,apoptosis rate,expression of Caspase-3 protein,level of ROS and content of MDA in curcumin group were decreased,while activity of SOD and level of miR-221 were increased(P<0.05).Compared with PD+miR-NC group,apoptosis rate,expression of Caspase-3 protein,level of ROS and content of MDA in PD+miR-221 group were decreased,and activity of SOD and level of miR-221 was increased(P<0.05).Compared with PD+anti-miR-NC+curcumin group,apoptosis rate,expression of Caspase-3 protein,ROS level and MDA content in PD+anti-miR-221+curcumin group were in-creased,while activity of SOD and level of miR-221 was decreased(P<0.05).Conclusion Curcumin can reduce 6-OHDA-in-duced oxidative damage in PD cell models by up-regulating miR-221.
