目的:探究黄贞菌质总黄酮(total flavonoids from Huangzhen mycoplasm,TFHM)的分离纯化工艺条件,并评估其对3种病原菌的抑制作用.方法:以吸附率、解吸率和总黄酮回收率为检测指标,从6种大孔树脂中筛选出最佳的树脂用于TFHM的分离纯化;采用单因素实验法考察不同工艺参数对TFHM分离纯化效果的影响;以滤纸片法检测经分离纯化后TFHM对3种病原菌的抑菌活性.结果:AB-8型大孔树脂为最佳分离纯化TFHM的树脂;在上样液浓度为5.4 mg/mL、上样液流速为1 mL/min和洗脱液流速为3 mL/min的条件下,TFHM的纯度达到了 54.47%,比初始样品提高了 26.27%;此外,经分离纯化后的黄贞菌质总黄酮对金黄色葡萄球菌和大肠杆菌均表现出较好的抑制作用,但对黑曲霉菌的抑制作用相对较弱.结论:AB-8型大孔树脂可以用于TFHM的分离纯化,且纯化工艺稳定可靠,所得的TFHM对金黄色葡萄球菌和大肠杆菌具有较好的抑制作用.
Isolation,Purification and Antibacterial Activity Analysis of Total Flavonoids from Huang Zhen Mycoplasm
Objective To explore the separation and purification process conditions of total flavonoids from Huang-zhen mycoplasm(TFHM)and evaluate their inhibitory effects on three pathogenic bacteria.Methods The best resin was selected from six kinds of macroporous resins for the separation and purification of TFHM.The single factor test method was used to investigate the influence of different process parameters on the separation and purification effect of TFHM.The filter paper method was used to detect the antibacterial activity of TFHM after separation and purifi-cation against three pathogenic bacteria.Results AB-8 macroporous resin is the best resin for separating and purif-ying TFHM.Under the conditions of a sample solution concentration of 5.4 mg/mL,a sample solution flow rate of 1 mL/min,and an eluent flow rate of 3 mL/min,the purity of TFHM reaches 54.47%,which is 26.27%higher than that of the initial sample.The TFHM after separation and purification shows good inhibitory effects on Staphylococcus aureus and Escherichia coli,but the inhibitory effect on Aspergillus niger is relatively weak.Conclusion:AB-8 macroporous resin can be used for the separation and purification of TFHM,and the purification process is stable and reliable.The obtained TFHM have good inhibitory effects on Staphylococcus aureus and Escherichia coli.
