摘要

为研究DEAD box蛋白5(DDX5)在头颈部鳞状细胞癌(HNSCC)中的表达水平及其对肿瘤发展和免疫细胞浸润的影响,利用UALCAN在线数据库对DDX5在HNSCC中的表达水平进行了深入分析,并探讨了其与HNSCC进展及HPV感染之间的潜在关联;通过CCK8实验,评估DDX5对HNSCC增殖能力的影响;应用在线数据库TIMER分析DDX5与HNSCC免疫细胞浸润的关系.结果表明:DDX5在HNSCC中显著上调且与HNSCC分化程度及HPV感染相关;DDX5能促进FaDu细胞的增殖能力;DDX5表达水平与CD4+T、树突状细胞(Den-dritic cells,DC)浸润水平呈正相关,且与Tregs细胞标记物呈显著正相关.综合上述分析可认为DDX5高表达促进HNSCC进展和免疫抑制,参与HNSCC发生发展.

Abstract

To investigate the expression level of DEAD box 5(DDX5)in head and neck squamous cell carcino-ma(HNSCC)and its effect on tumor progression and immune cell infiltration,the UALCAN online database was used to analyze the expression level of DDX5 in HNSCC,and the potential association between DDX5 and HNSCC progression and HPV infection was discussed.CCK8 assay was used to evaluate the effect of DDX5 on the prolifera-tion of HNSCC,and the online database TIMER was used to analyze the relationship between DDX5 and immune cell infiltration in HNSCC.The results showed that DDX5 was significantly up-regulated in HNSCC and correlated with the differentiation degree of HNSCC and HPV infection.DDX5 could promote the proliferation of FaDu cells.The expression level of DDX5 was positively correlated with the infiltration level of CD4+T cells and dendritic cells(DC),and was significantly positively correlated with Tregs cell markers.Based on the above analysis,it can be con-cluded that high expression of DDX5 promotes the malignant progression and immunosuppression of HNSCC,and participates in the occurrence and development of HNSCC.
