COMPARATIVE STUDY OF THE EFFECT OF DIFFERENT DOSES OF THE NEW COMBINED PROBIOTICS PREPARATION IN THE TREATMENT OF INTESTINAL BARRIER DAMAGE IN RATS WITH SEVERE BURNS
Objective To investigate the therapeutic effect of different doses of novel combined probiotic preparation for the treatment of intestinal barrier injury in severely burned rats.Methods 50%Ⅲ°burned rats model were established us-ing a 94℃water bath,and burned rats were intervened by three doses(0.5×109 CFU/d,1.5×109 CFU/d and 4.5×109 CFU/d)of probiotic preparations.ELISA,HE staining and intestinal barrier evaluation were used to observe the amelio-rative effects of different doses of probiotics on burns in rats,respectively.Results Compared with the sham group,the intestinal villi and crypt structure of rats in the burn group(Burn)were more severely damaged,and the height of intestinal villi was significantly reduced(P<0.05),whereas the probiotics treatment in the low-,medium-,and high-dose groups(P-L,P-M,and P-H,respectively)showed different degrees of improvement,among which the P-M group was the most effective group.Meanwhile,serum IL-1β,IL-6,and LPS levels were significantly reduced as well as IL-10 levels were significantly increased in the P-M group(P<0.05).Evaluation of the intestinal barrier function showed that the levels of diamine oxidase(DAO)and bacterial endotoxin in the small intestine of the P-L,P-M and P-H groups were significantly reduced com-pared with that of the Burn group(P<0.05),and the endotoxin levels in the P-M group showed the most significant reduction among the different dosages(P<0.05).Conclusions The combined probiotics preparation can significantly improve intes-tinal barrier damage in severely burned rats.The optimal dose was 1.5×109 CFU/d.
