EFFECTS OF SOLANINE ON BIOLOGICAL ACTIVITY,TUMOR INHIBITION AND MAPK/ERK1/2 IN LUNG CANCER A549 CELLS
Objective To investigate the effects of solanine on the biological activity,tumor inhibition and MAPK/ERK1/2 signal of A549 cells of lung cancer.Methods Human lung cancer cell line A549 was divided into blank group,low dose sola-nine group,medium dose solanine group,high dose solanine group and cisplatin group,and cultured with solanine concentra-tion of 0,15,20,25 μmol/L and 2 μg/mL cisplatin,respectively.Activity was measured by MTT;The apoptosis rate was detected by flow cytometry.Invasion was detected by Transwell method.Scratch test was performed to detect the scratch healing rate.25 nude mice were divided into blank group,low-dose solanine group,medium-dose solanine group,high-dose solanine group and cisplatin group.A549 cells were co-cultured with 0.3 mL of solanine 0,15,20,25 μmol/L and 2 μg/mL.Results The activ-ity of A549 cells was decreased,the apoptosis rate was increased,the invasion,scratch healing rate,MAPK,P-MAPK,ERK1/2 and P-ERK1/2 protein levels were decreased(P<0.05)in solasonine group.Conclusions Solasonine can inhibit the prolifera-tion,invasion and metastasis of lung cancer cells and accelerate apoptosis in a dose-dependent manner,with the best effect at high dose.The mechanism may be related to the inhibition of MAPK/ERK1/2 signal expression.
