摘要
保持冷冻后皮肤较好的活力对皮肤移植十分重要,采用慢速或中速降温(-1°C/min)或(-60°C/min)可以获得较高的活性.冷冻保护剂被证实对保持低温冻存皮肤活力有益,目前发现的冷冻保护剂有丙二醇、丙三醇、二甲基亚枫、海藻糖.采用海藻糖联合二甲基亚砜最有利于复合皮肤组织在深低温储存和冻融后保留活性.D-阿洛糖作为冷冻保护剂的添加剂,对皮肤角化细胞和人真皮成纤维细胞生存能力有益.玻璃化冷冻技术则使冻存后皮肤及皮肤相关组织、细胞活性达到80%以上.
Abstract
Objective: Keeping better skin viability after freezing is very important for skin transplantation. higher viabilities were obtained when using a slow (-1 degree C min-1) or medium (-60 degree C min-1) freezing. At present, the discovered of cryoprotectants are dimethyl sulfoxide, ethanediol, glycerol and trehalose. The use of trehalose, in combination with dimethyl sulfoxide (DMSO) in the cryopreservation is benefit for a retention of skin tissue flaps activity after freezing thawing. D-allose, a sole additive, may provide effective protection for human skin keratinocytes and dermal fibroblasts cells during freezing. The vitrification technique protect over 80 % viability of skin tissue and Relation tissue of skin and cell.
