摘要

目的 采用网络药理学研究复方龙血竭胶囊治疗心绞痛的潜在分子机制.方法 通过TCMSP数据库、BATMAN-TCM数据库并结合国内外文献调研获取复方龙血竭胶囊组成药材龙血竭、三七、冰片的主要化学成分.通过DrugBank、SwissTargetPrediction数据库获取复方龙血竭胶囊化学成分的作用靶点.利用GeneCards、OMIM和TTD数据库检索"angina pectoris"相关靶点,得到复方龙血竭胶囊和心绞痛交集靶点.应用STRING数据库Cytoscape 3.7.2软件构建蛋白相互作用(PPI)网络及"复方龙血竭胶囊-活性成分-关键作用靶点"网络,并对该网络进行拓扑学分析.利用R软件对复方龙血竭胶囊的交集靶点进行GO富集分析和KEGG通路富集分析,预测其作用机制.结果 复方龙血竭胶囊包含活性成分107个,对应靶标568个,其中与心绞痛相关的共同靶标基因有50个;涉及1033个生物过程,62个分子功能,39个细胞组成,13条信号通路.结论 复方龙血竭胶囊可通过IL6、TNF、VEGFA、NOS3等多靶点,调节低氧诱导因子1(HIF-1)、NF-κB信号通路、MAPK信号通路、PI3K/AKT信号通路、钙信号通路等多途径发挥治疗心绞痛的作用.

Abstract

Objective To explore the possible mechanism of Fufanglongxuejie Capsules in treating angina pectoris by network pharmacology.Methods The related chemical components of three Chinese medicines in Fufanglongxuejie Capsules were collected by TCMSP database,BATMAN-TCM database,and related litera-tures by Pub Med and CNKI.DrugBank,SwissTargetPrediction were used to obtain the targets of Fufang-longxuejie Capsules.GeneCards,OMIM and TTD databases were used to search the potential targets of"angina pectoris",and then the core targets of Fufanglongxuejie Capsules and angina pectoris targets were obtained.A protein-protein interaction(PPI)network and the"Fufanglongxuejie Capsules composition-components-tar-gets"network were constructed by STRING database and Cytoscape 3.7.2 software.GO analysis and KEGG pathway analysis were performed by using R software.Results Fufanglongxuejie Capsules contained 107 active components,568 targets,and 50 potential targets with angina pectoris were obtained.GO analysis and KEGG analysis showed that they involved 1033 biological processes,62 molecular functions,39 cellular components,and 13 signaling pathways.Conclusion Fufanglongxuejie Capsules can treat angina pectoris through multi-components,multi-targets and multi-pathways interaction,which provides a new direction for the basic and clinical research of Fufanglongxuejie Capsules in the treatment of angina pectoris.
