Objective To investigate the effect and mechanism of dexmedetomidine(Dex)on JAK2/STAT3 signaling pathway in interleukin-6(IL-6)-induced inflammatory alveolar epithelial cell model.Methods Inflammatory alveolar epithelial cell model was constructed by treating MLE12 cells with IL-6,and the cells were divided into five groups by random number method:the blank control group(group C)was cul-tured without intervention in culture medium for 96 h;the IL-6 group(group I)was added with 50 ng/ml IL-6 into the culture medium;and the IL-6+dexmedetomidine group(group I+D)was added with 50 ng/ml IL-6 into the culture medium,and after 24 h,10 μMM DEX was added to the culture medium;IL-6+HY-121488 group(Group I+H)added 50 ng/ml IL-6 to the culture medium after adding 50 μ.M HY-121488;IL-6+HY-121488+DEX group(Group I+H+D)after adding 50 μM HY-121488 to the medium,50 ng/ml IL-6 was added to the medium and 10 μM DEX to the medium 24 h later.Cells in each group were col-lected for experimental detection after 72 h.The mRNA expression levels of IL-6,JAK2 and STAT3 were de-tected by PCR.Results Compared with group C,the mRNA expression levels of IL-6,JAK2,and STAT3 mRNA were increased in group I(P<0.05);compared with group I,the mRNA expression levels of IL-6,JAK2,and STAT3 mRNA were decreased in group I+D(P<0.05);and compared with group I+D,the mRNA expression levels of IL-6,JAK2,and STAT3 mRNA were increased in group I+H+D(P<0.05);compared with group I,IL-6,JAK2,and STAT3 mRNA expression levels were all increased in group I+H(P<0.05);compared with group I+H,IL-6,JAK2,and STAT3 mRNA expression levels were all decreased in group I+H+D(P<0.05).Conclusion Dexmedetomidine attenuates the inflammatory response and exerts a protective effect on lung tissues by modulating the gp130-mediated JAK2/STAT3 signaling pathway.
