摘要
目的 本研究通过16S rRNA基因测序技术,探究人乳头瘤病毒(human papillomavirus,HPV)感染患者与HPV阴性健康女性阴道微生态变化以及菌群功能差异,为临床治疗提供理论基础.方法 选取2023年3-12月于本院妇科门诊就诊的女性242例,将HPV阳性样本设为HPV感染组(n=97),HPV阴性样本设为Control组(n=145).将两组样本分别进行16S rRNA测序后通过生信分析研究两组样本的Alpha多样性、Beta多样性、群落组成结构,进而探究阴道微生态与HPV感染的相关性.结果 Alpha多样性与Beta多样性分析结果表明,Control组阴道菌群的丰富度和多样性显著高于HPV感染组,且两组阴道菌群分布组成具有显著差异.堆叠图结果表明与Control组相比,HPV感染组阴道菌群结构发生明显变化.结论 HPV感染后患者的阴道菌群多样性和丰富度显著降低,尤其是乳酸杆菌属、厚壁菌门、梭杆菌属、变形菌门等菌群显著降低,并且两组阴道菌群的分布组成存在显著差异.
Abstract
Objective In this study,16S rRNA gene sequencing technology was used to explore the changes of vaginal microecology and the difference of flora function between patients with human papillomavirus(HPV)infection and HPV-negative healthy women,so as to provide theoretical basis for clinical treatment.Method A total of 242 female patients were selected from the gynecological clinic of a hospital from March 2023 to December 2023.The HPV positive samples were set as the experimental group(HPV,n=97),and the HPV negative samples were set as the control group(Control,n=145).The two groups of samples were se-quenced by 16S rRNA,and the Alpha diversity,Beta diversity and community composition structure of the two groups of samples were studied by bioinformatics analysis,so as to explore the correlation between vaginal mi-croecology and HPV infection.Results The results of Alpha diversity and Beta diversity analysis showed that the richness and diversity of vaginal flora in the Control group were significantly higher than those in the HPV infection group,and the distribution of vaginal flora in the two groups was significantly different.The results of the stack diagram showed that compared with the HPV negative control group,the vaginal flora structure of the HPV infection group changed significantly.Conclusion The diversity and richness of vaginal flora in patients with HPV infection were significantly reduced,especially Lactobacillus,Firmicutes,Fusobacterium,Proteobac-teria and other genera were significantly reduced,and there were significant differences in the distribution of vaginal flora between the two groups.
