基于生物信息学的高尿酸血症伴缺血性脑卒中的关键基因及中药预测

Key Genes and Traditional Chinese Medicine Prediction Based on Hyperuricemia with Ischemic Stroke Based on Bioinformatics

目的 通过生信分析筛选缺血性脑卒中(IS)合并高尿酸血症(HUA)的关键基因,并预测中药.方法 下载基因表达综合数据库(GEO)中HUA和IS的微阵列数据.使用R包鉴定与HUA和IS相关的共表达基因.使用DAVID对HUA和IS中存在的共享基因进行富集分析和生物过程分析,并使用Cyto-cape筛选出其关键基因,并进行验证.最后利用8个关键基因进行映射,筛选中药姜黄、丹参、黄芩.结果 将51个基因确定为HUA和IS的共享基因并进行富集,发现炎症反应是HUA和IS病理生理学的共同特征.并根据HUA和IS中共享和关键基因特征,映射其对应的潜在中药.结论 采用生信分析进行HUA合并IS的关键基因探索,并确定可用作生物标志物或潜在治疗靶点的新型候选基因和中药预测分析.
Objective To screen the key genes of ischaemic stroke(IS)combined with hyperuricemia(HUA)by raw letter analysis and to predict Chinese medicine.Methods Microarray data of HUA and IS were downloaded from the Gene Expression Omnibus(GEO)database.R packages were identified for co-expressed expressed genes associated with HUA and IS.The shared genes present in HUA and IS were enriched and anal-ysed for biological processes using DAVID,and their key genes were screened and validated using cytocape.Fi-nally,eight key genes were used for mapping to screen the Chinese medicines turmeric,danshen and scutellaria.Results Fifty-one genes were identified and enriched as shared genes of HUA and IS,and inflammatory re-sponse was found to be a common feature of HUA and IS pathophysiology.And based on the shared and key gene features in HUA and IS,their corresponding potential herbal medicines were mapped.Conclusion Biosense was used for key gene exploration of HUA combined with IS and to identify novel candidate genes that can be used as biomarkers or potential therapeutic targets and herbal medicine predictive analyses.
