目的:探讨微小RNA-22-5p(microRNA-22-5P,miR-22-5p)在乳腺癌患者外周血中的表达及其与肿瘤相关性.方法:选取2017年6月-2018年5月于海安市人民医院肿瘤科就诊的成年乳腺癌患者60例及同期健康体检者30名,分别为乳腺癌组及健康对照组.应用实时定量聚合酶链式反应(quantitative real time polymerase chain reaction,qRT-PCR)检测两组研究对象外周血miR-22-5p水平;绘制ROC曲线评估miR-22-5p对乳腺癌的诊断效能;采用Kaplan-Meier生存曲线分析外周血miR-22-5p水平对乳腺癌的预后价值.结果:qRT-PCR结果显示,乳腺癌组患者外周血miR-22-5p相对表达量显著低于健康对照组(P<0.05);乳腺癌不同分子分型、TNM分期、淋巴结转移、远处转移、脉管受侵、组织学分级之间miR-22-5p水平比较差异有统计学意义(P<0.05);外周血miR-22-5p诊断乳腺癌的AUC为0.882,CA153诊断乳腺癌的AUC为0.667,联合检测的AUC为0.901;Kaplan-Meier生存分析显示,miR-22-5p低水平组患者5年总生存期明显高于miR-22-5p高水平组(P<0.05).结论:MiR-22-5p在乳腺癌的诊断及预后评估方面有一定的临床价值.
Diagnosis and prognostic value of peripheral blood miR-22-5p expression in breast cancer patients
Objective:To investigate the expression of microRNA-22-5p(miR-22-5p)in peripheral blood of breast cancer patients and its correlation with tumor.Methods:Sixty adult breast cancer patients and 30 healthy individuals who underwent medical examinations from June 2017 to May 2018 at Department of Oncology in Haian People's Hospital were selected as the breast cancer group and the healthy control group,respectively.Quantitative real time polymerase chain reaction(qRT-PCR)was used to detect the levels of peripheral blood in both groups.ROC curves were plotted to evaluate the diagnostic efficacy of miR-22-5p for breast cancer.Kaplan-Meier survival curves were used to analyze the prognostic value of peripheral blood miR-22-5p levels in breast cancer.Results:qRT-PCR results showed that the relative expression level of miR-22-5p in pe-ripheral blood of breast cancer patients was significantly lower than that in the healthy control group,with statistical signifi-cance(P<0.05).The comparison of miR-22-5p levels between different molecular subtypes,TNM stages,lymph node metastasis,distant metastasis,vascular invasion,and histological grades of breast cancer showed significant differences(P<0.05).The AUC of peripheral blood miR-22-5p for diagnosing breast cancer was 0.882,while that of CA153 was 0.667.The combined detection had an AUC of 0.901.Kaplan-Meier survival analysis showed that patients with low levels of miR-22-5p had significantly higher 5-year overall survival compared to those with high levels of miR-22-5p(P<0.05).Conclusion:MiR-22-5p has certain clinical value in the diagnosis and prognosis evaluation of breast cancer.
