Experimental study on irritation and allergy of Tuihuangsan
Objective:To study the skin irritation and allergic reactions of Tuihuangsan and to provide a toxicological basis for its use in clinical practice.Methods:Skin stimulation experiment in rabbits:4 rabbits,male and female,were used.The skin of both sides of the spine of rabbits was prepared(3 cm×3 cm)by the self-control method,and the left side was adminis-tered with Tuihuangsan and the right side was administered with warm water once a day,and the drug was repeatedly admin-istered for 14 days,and the skin was observed for erythema and edema,and the skin was scored for irritation and intensity.Thirty guinea pigs,half male and half female,were selected and divided into a blank control group(warm water),a positive control group(2,4-dinitrochlorobenzene:sensitising concentration of 1%,excitation concentration of 0.1%)and Tuihuangsan group,with 10 guinea pigs in each group.The guinea pigs were sensitized on the left side of the skin preparation area on days 1,7 and 14,and the right side of the skin preparation area was stimulated on day 14 after the last sensitization,and the guinea pigs were observed for erythema,edema and other allergic reactions at 1 h,24 h,48 h and 72 h after the stimulation,and the mean values of reactions and sensitization rates were calculated for each group.Results:The skin of rabbits was not irritated by the antireflux powder,and no histopathological changes were observed in the pathological examination;in the guinea pig skin sensitization test,no erythema and edema were observed in the skin,and the sensitization rate was 0%.Conclusion:Tui-huangsan is non-irritating to the skin of rabbits and non-allergenic to the skin of guinea pigs,which is of good safety and can be promoted for use in the clinic.
