Research progress on the mechanism of inhibition of ovarian cancer with metformin based on old drugs and new use
Among gynecological malignant tumors,ovarian cancer has the highest fatality rate,because the initial clinical symptoms of ovarian cancer are not obvious,most patients are in the terminal stage of disease development at the time of pre-sentation,often accompanied by abdominal and pelvic effusion and metastasis of cancer cells,etc.,while ovarian cancer cells are prone to resistance to platinum-based chemotherapy drugs,and relapsed resistant patients currently have no unified treat-ment plan,drug-resistant cancer cells to carboplatin,paclitaxel,second-line chemotherapy drugs(docetaxel,Doxorubicin lipo-somes)and so on have no obvious reaction,so the fatality rate of ovarian cancer has remained high.In order to improve the treatment effect of ovarian cancer and delay the recurrence,it is necessary to research new treatment methods.Compared with the study of new drugs,mining the anti-tumor effect of traditional non-tumor drugs may be a safe and efficient research idea.Among them,metformin is a typical representative of"old drugs and new use",a derivative of metformin,the main indication is type 2 diabetes.Recently,with the in-depth study of the mechanism of action and biological activity of metformin,it has been found that it has the potential for anti-cancer effects.Metformin has inhibitory effects on breast cancer,ovarian cancer,pancreatic cancer,lung cancer and other malignant tumors.Its main mechanisms include AMP activated protein kinase path-way,improvement of platinum resistance,inhibition of cancer cell metastasis,change of immune cell function,sensitivity,etc.This article reviews the specific mechanism of metformin in inhibiting ovarian cancer.
