Establishment of drug sensitivity model of Sorafenib in hepatocellular carcinoma cell lines with different metastatic potential
Objective:To investigate the drug sensitivity of Sorafenib in hepatocellular carcinoma(HCC)cell lines with different metastatic potential and in HCC xenografts generated in nude mice and to establish a drug sensitivity model of Sorafenib in HCC.Methods:The drug sensitivity of Sorafenib in HCC cell lines SMMC-7721(low-metastatic potential)and MHCC-97H(high-metastatic potential)and corresponding HCC xenografts generated in nude mice was detected by ATP anti-cancer drug sensi-tivity assay.Results:According to the sensitivity criterion,the IC50 value of Sorafenib in SMMC-7721 cell line group was sig-nificantly lower than that in MHCC-97H cell line group.Sorafenib was highly sensitive in SMMC-7721 xenograft group and moderately sensitive in MHCC-97H xenograft group.The chemotherapy sensibility index(CSI)and AUC values of Sorafenib in SMMC-7721 cell line group and the corresponding xenograft group were both better than those in MHCC-97H cell line and the corresponding xenograft groups,respectively.The growth inhibition rate of Sorafenib in SMMC-7721 cell line and corre-sponding xenograft groups was higher than that in MHCC-97H groups.Conclusion:Sorafenib exhibited varying drug sensitivity,CSI and AUC values,as well as growth inhibition rates in the SMMC-7721 cell line with low-metastatic potential and the MHCC-97H cell line with high-metastatic potential,particularly evident in the respective xenograft models established in nude mice.The drug sensitivity model of Sorafenib in HCC cell lines with different metastatic potential was successfully established.
