丽格海棠叶片优化培养高效再生体系的研究

以丽格海棠叶片为外植体,进行不定芽的诱导和植株再生试验研究,结果表明:叶切块可直接分化不定芽,以MS+6-BA2.0 mg/L+TDZ0.1 mg/L+NAA0.5 mg/L是较适合的分化培养基,分化率达100%,平均芽数8.1;不定芽增殖以MS+6-BA 1.0 mg/L+KT0.5 mg/L+NAA0.2 mg/L效果较好,平均增殖倍数为12.2左右,生根培养基以1/2MS+NAA0.1 mg/L+IBA0.3 mg/L效果较好,平均生根数为4.4条左右,生根率为100%;瓶苗移栽于森林土∶草碳=(1∶1)的基质中生长良好,成活率达98%以上.
Research of optimization of culture and high efficient regeneration system for Begonia × Elatior(balamon) using leaf blade
The leaf blade ofBegonia×Elatior(balamon)was used as explants to research on adventitious bud induction and plantlet regeneration. The results showed that:cut of Leaves can be directly differentiation of adventitious buds the suitable differential medium for adventitious buds induction was MS+6-BA2.0 mg·L-1+TDZ0.1 mg·L-1+NAA0.5 mg·L-1 the differentiation rate and mean number of shoots could reach to 100%and8.1; it could effectively improve the proliferation of adventitious buds for MS+6-BA 1.0 mg·L-1+KT0.5 mg·L-1+NAA0.2 mg·L-1and obtained a higher proliferation rate of 12.2, the optimal rooting medium was 1/2MS+NAA0.1mg·L-1+IBA0.3mg·L-1 with its average survival rate and root number up to100%,4.4 respectively; the test-tube plantlets were easy to be transplanted and survive when it transplanting medium was forest soils∶Substratum=1∶1 , and showed 98%survival rate.
