灵武长枣叶阿拉伯半乳糖蛋白分布及光合特性

以灵武长枣4个发育时期的叶为试验材料,利用免疫荧光定位方法,研究不同发育时期的叶中阿拉伯半乳糖蛋白(AGPs)的分布特征,并测定叶的光合特性及可溶性糖质量比,以期揭示灵武长枣不同发育时期叶的AGPs分布特征、光合特性及干旱适应特征.结果表明:①不同时期的叶表皮细胞外切向壁具有较厚的角质层,抗体所识别的抗原荧光AGPs分布较多;叶肉全部由发达的栅栏组织细胞组成,不同发育时期叶肉细胞壁和细胞内部均密集分布着抗体所识别的抗原荧光AGPs,是叶AGPs分布的主要部位,且后期分布增强.不同时期主脉以及位于叶肉中的侧脉和细脉维管束木质部和韧皮部细胞壁和细胞内部均分布着大量抗体所识别的抗原荧光AGPs,维管束鞘和薄壁组织中的分泌道内均没有抗原荧光AGPs分布.②叶的净光合速率与气孔导度呈高低交替的变化趋势,在花后90 d达到最高或较高水平,蒸腾速率在花后60 d达到最高值后逐渐降低,胞间CO2浓度与蒸腾速率的变化趋势相反.葡萄糖和果糖质量比的变化相似,为逐渐上升的趋势,且葡萄糖的质量比始终高于果糖;蔗糖质量比随着叶的发育呈先下降后逐渐上升达到峰值的趋势,蔗糖质量比的增长速度和增长量始终远高于葡萄糖和果糖,表明叶片以积累蔗糖为主.叶的光合特性及可溶性糖质量比的变化具有明显规律.可见,在叶的不同组织中抗体所识别的抗原荧光强弱存在一定差异,叶肉始终是叶AGPs分布的主要部位;AGPs可能参与了保护、抵御干旱、营养支持等.
Distribution of Arabinogalactan Proteins and Photosynthesis Characteristics in Ziziphus jujuba Mill'Lingwuchangzao'Leaf
To reveal the distribution of arabinogalactan proteins AGPs and photosynthesis characteristics and drought adaptation characteristics in different development periods leaf of Ziziphus jujuba Mill'Lingwuchangzao',leaves of four periods were used as experimental material to study the distribution of AGPs by methods of immunofluorescence localization,and the photosynthesis characteristics and the soluble suger contents index of leaf were determined.The results showed that:① the external tangential cell walls of leaf epidermis cells in different developmental stages formed quite thick cuticles,antigen fluorescence of AGPs recognized by antibody distributed more copious.The mesophyll consisted entirely of developed palisade tissue cells,the cell walls and cell interiors of the mesophyll distributed antigen fluorescence of AGPs recognized by antibody in different periods leaf,the mesophyll was the main tissue of leaf AGPs distribution and the later distribution was enhanced.The cell walls and cell interiors of xylem and phloem of vascular bundles in main veins,lateral veins and veinlets located in the mesophyll distributed mass antigen fluorescence of AGPs recognized by antibody in different development periods,there were no antigen fluorescence of AGPs recognized by antibody in the vascular bundle sheaths and the secretory ducts in parenchyma.② The Pn and Gs of leaves showed a trend of alternating high and low,and reached the highest or higher level at 90 days after blossom;the Tr reached the highest value at 60 days after blossom and then decreased gradually;and the change trend of Ci was opposite to that of Tr.The contents of glucose and fructose showed a similar trend of gradual increase,and the content of glucose was always higher than that of fructose.The contents of sucrose decreased first and then gradually increased to the highest with the development of leaves,the increase speed and increment of sucrose content were always much higher than those of glucose and fructose,indicating that sucrose was mainly accumulated in leaves.The photosynthesis characteristics and the soluble suger contents of leaf showed obvious variations laws.All the results above indicated that there were some differences in fluorescence intensity of antigen recognized by antibody in different leaf tissues,the mesophyll was the main tissue of AGPs distribution in leaf all along,and AGPs probably participated in protection,drought resistance and nutritional support.
