Study on the Hypoglycemic Activity of Potato Starch and Starch Products from Guyuan in Diabetic Rats
Guyuan City is one of the four major potato planting bases in China.Due to natural environmental factors such as high altitude,concentrated rainfall during the growing season,and significant temperature differences between day and night,Guyuan is highly suitable for high-quality potato production.To investigate the hypoglycemic effects of Guyuan potato starch,this study established a novel method for determining blood glucose levels using 1-phenyl-3-methyl-5-pyrazolone(PMP)pre-column derivatization high-performance liquid chromatography(HPLC).This method demonstrated good linearity within the measurement range,with the accuracy of glucose reference solutions at three quality control concentrations ranging from 89.7%to 110.4%.The relative standard deviation(RSD)was less than 6.5%,and the recovery rate of plasma samples was between 81%and 103%.Validation results indicated that this method is simple,accurate,stable,and suitable for determining blood glucose levels in rats.Furthermore,this study,for the first time,measured the content of slowly digestible starch(SDS)and resistant starch(RS)in different starch types by combining in vitro simulated digestion with in vivo animal model evaluation.The hypoglycemic properties of Guyuan potato starch and its starch products in diabetic rats were confirmed,providing a scientific basis for the application of Guyuan potatoes in the field of food nutrition and health.
