昌邑湿地可培养细菌多样性分析及产酶菌株筛选

分析昌邑湿地环境可培养细菌物种多样性及其产酶潜力.采用10倍梯度稀释涂布平板的培养方法,从昌邑湿地土壤中分离可培养细菌,通过16S rRNA基因序列分析昌邑地区可培养细菌的多样性;同时对菌株进行了生物学活性及耐盐性测定.根据16S rRNA基因序列的系统进化分析,将分离得到的93株细菌分别归类到3个门的7个属,其中芽孢杆菌属(Bacillus)为该湿地土壤可培养细菌的优势菌群,占所有菌株的50%.细菌酶活检测结果表明:60株细菌具有纤维素酶活性,51株细菌具有蛋白酶活性,50株细菌具有淀粉酶活性,其中34株细菌同时具有3种酶活性.研究表明昌邑湿地土壤微生物具有丰富的多样性,并获得了产酶活性良好的耐盐菌株资源,为研究滨海湿地在环境治理及净化能力方面提供基础参考.
In order to analyze the biodiversity and enzyme activity of cultivable bacteria in soils of Changyi National Marine Ecological Special Reserve.93 strains were isolated by 10-fold serial dilution method,and identified by 16S rRNA genes.Enzyme activities(including amylase,protease and cellulase)and NaCl tolerance were detected.Phylogenetic analysis based on 16S rRNA showed that the 93 bacteria belonged to 3 classes and 7 genera.The pre-dominant genera were Bacillus,comprising 50%of all bacteria.The results of enzyme ac-tivity test shows that the number of strains with cellulase activity,protease activity and am-ylase activity are 60,51 and 50,respectively.What is more,34 strains of them had 3 enzyme activi-ties.This study showed that the biodiversity of cultivable bacteria in soil of Changyi wetland,and some salt-tolerant strains with enzyme activities were obtained.Those provided a basis refer-ence for the study of environmental governance and purification capacity in coastal wetland.
