Preparation,Quality and in Vitro Activity of Dispersible Tablets of Aronia melanocarpa
The process for the preparation of Aronia melanocarpa dispersible tablets and their in vitro activity were explored.A combination of one-way and orthogonal tests was used to examine the prepa-ration process, the content and in vitro activity of total triterpenes, and to determine the content of gal-lic acid, catechin, and epicatechin in the A.melanocarpa dispersible tablets by ultra-high-perform-ance liquid chromatography.Microcrystalline cellulose as diluent, crosslinked polyvinylpyrrolidone as disintegrant,75%ethanol as binder and micronised silica gel as lubricant were the preferred prescrip-tions for the A.melanocarpa dispersible tablets. The method for the determination of total triterpenes in the dispersible tablets showed that the stability before and after colour development was in accordance with the requirements of the assay, the precision and reproducibility of the instrument were good, and the recoveries of the total triterpenes were 99.48%.The results of the in vitro activity test showed that the Aronia melanocarpa dispersible tablets had the ability to scavenge DPPH and inhibit liposomes. The dispersible tablets prepared by the preferred formulation process have a disintegration time limit in accordance with the requirements and a total triterpene content of 1.42 mg/tablet.Ultra-high perform-ance liquid chromatography(UPLC)was performed on an Acquity Uplc Hss C18 column with metha-nol(A)-0.2%aqueous phosphoric acid(B)as the mobile phase in a gradient elution. The recoveries of orotate, catechin and epicatechin were 99.3%at the detection wavelength of 280nm.The spiked re-coveries of gallic acid, catechin, and epicatechin were 99.83%,100.69%,and 99.61%,respective-ly.The contents of the three components were determined and analysed as 111.15 pg/g,940.19μg/g and 598.25 µg/g,respectively.The results of the test provide a basis for the application of the A.melanocarpa dispersible tablets.
