Study and Establishment of TLC Identification Method of Eupatorium chinense in Guizhou Province
In order to establish a thin-layer chromatography(TLC)identification method for Eupatori-um chinense Linn.produced in Guizhou province,a methodological study was conducted using Quer-cetin as the standard substance,following the TLC method(General Rule 0502).Factors such as de-veloping agent,sample size,extraction method,extraction solvent,chromogenic agent,inspection wavelength,silica gel G plates from different manufacturers,and environmental conditions(humidity and temperature)were investigated.The results showed that the best method for TLC identification of E.chinense medicinal herb is:the sample size of the test sample is 5 µL.The standard sample size is 2 μL.The developing agent is toluene ethyl acetate formic acid(5∶2∶1),the color developing a-gent is aluminum chloride ethanol solution of 3%,the color developing temperature is 105℃,and the inspection wavelength is 365 nm.In addition,research has found that the extraction solvent,extraction method,silica gel G plates of different manufacturers,and environmental conditions(humidity and temperature)have little effect on the identification of TLC.Overall,this method is simple to operate,has strong specificity,good stability and reproducibility,and can be used for the qualitative identifica-tion of Guizhou produced Eupatorium chinense Linn by TLC.
