EXPRESSION OF HYPOXIA INDUCIBLE FACTOR-2α IN PLACENTA OF NORMAL PREGNANT WOMEN AND OF PATIENTS WITH PREECLAMPSIA
Objective To understand the expression of hypoxia inducible factor-2α (HIF-2α) in placenta of normal pregnant women and of patients with preeclampsia as well as its role in pathogenesy of preeclampsia.Methods Semi-quantitative RT-PCR and immunohistochemistry were used to detect the expressions of HIF-2α mRNA and protein in placenta of 30 patients with preeclampsia, in which, 11 mild and 19 severe, and 37 normal pregnancies served as controls, of whom, 12 early, 10 midtrimester, and 15 late pregnancies.Results In normal pregnancy: the expression of HIF-2α mRNA in placenta of the early pregnancy was lower than that of the late pregnancy (F= 5.323, q=4.574, P<0.01 ), but the differences were not significant between the other groups (P>0.05).The mean density of HIF-2α protein in the early group was higher than that in both the midtrimester and the late group, the difference being significant (F=11.862tq=6.230,5.672;P<0.01), but the difference of that between the mid and late group was not significant (P>0.05).In preeclampsia, HIF-2α protein in placenta was higher than that in the late pregnancy (t= 5.174, P<0.01), and the difference of that between severe and mild groups was not significant (P>0.05).Conclusion HIF-2α plays a role in the development of placenta and involves in the pathophysiological processes of preeclampsia.
