Objective To investigate the polymorphism of the BALF2 gene and its distribution characteristics and its rela-tionship with the development and progression of Epstein-Barr virus(EBV)-associated tumors. Methods DNA sequence analysis was performed on 349 EBV-positive specimens using nested PCR and DNA sequencing.Lasergene software was used for sequence alignment of them with the standard EBV strain B95-8 to derive a phylogenetic tree for classification of gene variants. Results According to the phylogenetic tree, there were five BALF2 genotypes:BALF2-A,BALF2-B,BALF2-C,BALF2-D,and BALF2-E.The detection rate of BALF2-E was higher in people with nasopharyngeal carcinoma than in healthy people, and higher in southern people with nasopharyngeal carcinoma than in northern people with this disease in China(x2=10.26,P<0.01).Con-clusion The BALF2 gene is polymorphic, and the distribution of its variants is associated with tumor types and geographic re-gions.The BALF2-E variant is related to the development of nasopharyngeal carcinoma, especially in the high-incidence areas.
