Objective To investigate the effect of ferric ammonium citrate(FAC)on the electrical activity of dopaminergic neurons in the substantia nigra of mice.Methods Brain slices were prepared from the C57BL/6 mice of 15-20 days of postnatal age,and the whole-cell patch clamp technique was used to observe the effect of FAC on the spontaneous discharge and membrane potential of substantia nigra dopaminergic neurons.Results After the brain slices of the substantia nigra of mice infiltrated with 100 μmol/L FAC solution for 5 minutes,there was a significant reduction in the frequency of spontaneous discharge of dopaminer-gic neurons(t=5.533,P<0.01),and after 10 minutes,there was a significant reduction in the frequency of spontaneous discharge of dopaminergic neurons(t=7.297,P<0.001).The neuronal membrane potential moved towards hyperpolarization(t=4.227,P<0.01).Conclusion FAC can inhibit the electrical activity of dopaminergic neurons in the substantia nigra of normal C57BL/6 mice.
