不同时间ARS对雄性CD1小鼠运动及情绪影响

摘要
目的 探讨不同时间急性束缚应激(ARS)对雄性CD1小鼠运动、焦虑及抑郁样行为的影响.方法 将8周龄CD1雄性小鼠随机分为对照组、ARS 0.5 h组、ARS 1.0 h组、ARS 2.0 h组和ARS 4.0 h组,每组10只.造模后分别进行糖水偏好实验、旷场实验和高架十字迷宫实验.结果 糖水偏好实验结果显示,与对照组相比,ARS各组小鼠蔗糖偏好百分比无明显差异.旷场实验结果显示,与对照组相比,ARS各组小鼠总移动距离和运动速度无明显差异.高架十字迷宫实验结果显示,与对照组相比,ARS 0.5 h组、ARS 1.0 h组和ARS 2.0 h组小鼠进入开放臂的次数显著减少(q=4.483~6.150,P<0.01),在开放臂中停留时间明显减少(q=3.607~4.665,P<0.05);ARS 4.0 h组小鼠进入到开放臂中的时间和次数与对照组相比无明显差异.结论 不同时间ARS不影响CD1小鼠的抑郁样行为及运动能力,但ARS0.5、1.0和2.0 h能诱导CD1小鼠产生焦虑样行为.
Abstract
Objective To investigate the influence of different durations of acute restraint stress(ARS)on motor ability,anxiety,and depression-like behavior in male CD1 mice.Methods Male CD1 mice aged 8 weeks were randomly divided into con-trol group,0.5 h ARS group,1.0 h ARS group,2.0 h ARS group,and 4.0 h ARS group,with 10 mice in each group,and the su-crose preference test,the open field test,and the elevated plus maze test were performed after modeling.Results The sucrose preference test showed that there was no significant difference in the percentage of sucrose preference between the ARS groups and the control group.The open field test showed that there were no significant differences in total movement distance and movement speed between the ARS groups and the control group.The elevated plus maze testshowed that compared with the control group,the 0.5 h ARS group,the 1.0 h ARS group,and the 2.0 h ARS group had significant reductions in the number of times of entering the open arm(q=4.483-6.150,P<0.01)and the time spent in the open arm(q=3.607-4.665,P<0.05),while there were no significant differences in these two indicators between the 4.0 h ARS group and the control group.Conclusion Different dura-tions of ARS do not affect the depression-like behavior and motor ability of CD1 mice,but ARS for 0.5,1.0,and 2.0 h can induce anxiety-like behavior in CD1 mice.
