Expressions of HIF-1α,VEGF and HK2 in gastric cancer cells inhibited by muscone under hypoxia
Objective To investigate the effects of muscone on the mRNA and protein expressions of hy-poxia-inducible factor 1(HIF-1α),glycolytic enzyme hexokinase-2(HK2)and vascular endothelial growth factor(VEGF)in gastric cancer cells under hypoxia.Methods Expressions of HIF-1α,HK2 and VEGF in STAD(Stomach adenocarcinoma)and paracancerous tissues of gastric cancer were analyzed by GEPIA data-base.The relationship between gene expression and prognosis of gastric cancer was analysed using Log-rank and Breslow test on KM-plotter website.The expressions of three genes were detected using quantitative real-time fluorescence PCR(qRT-PCR).Protein blotting(Western Blot)was adopted to analyse the effects of mus-cone on the expressions of HIF-1α,VEGF and HK2 in gastric cancer cells HGC27 under hypoxia(formed by CoCl2).Results HIF-1α and HK2 were highly expressed in gastric cancer tissues(HIF-1α,HK2;P<0.01).The prognostic assessment suggested that VEGF was a gastric cancer risk gene(P<0.05),which was associat-ed with poor prognosis of gastric cancer patients.The results of qRT-PCR showed that the mRNA expression of VEGF was down-regulated after 8 h of muscone treatment on HGC27 cells under hypoxia.The mRNA expres-sions of HIF-1α and HK2 were down-regulated after 24 h.Protein blotting showed that HIF-1α protein ex-pression was reduced after muscone treatment on HGC27 cells under hypoxia for 24 h(P<0.05).Conclusion Muscone may exert inhibitory effects on the mRNA and protein expressions of HIF-1 α,HK2 and VEGF un-der hypoxia.
