EPSTEIN-BARR VIRUS EBNA2 POLYMORPHIC PATTERNS IN NASOPHARYNGEAL CARCINOMA BETWEEN POPULATION IN QINGDAO AND GUANGZHOU
Objective To compare Epstein-Barr virus (EBV) nuclear antigen 2(EBNA2) encoding gene polymorphism in EBV positive nasopharyngeal carcinoma (NPC) tissue in crowd of Qingdao and Guangzhou and explore the correlation between EBV gene subtypes and NPC.Methods Tissues of EBV positive NPC were taken from 32 patients,and throat washing (TW) samples collected from 36 EBV positive healthy individuals of Guangzhou.Employing PCR technique,amplification and sequencing were done for EBNA2 aa331-487 gene.Results The subtype E2 A was the main type of EBNA2 in Guangzhou.In NPC tissue,the distribution of EBNA 2 subtypes was higher than that in TW of healthy adults,the difference was significant (x2 =12.2,P <0.01),and E2 A subtype was also a main type found in Qingdao,but the detection rate of E2 A in NPC tissue was much higher in Guangzhou versus that in Qingdao (x2 =18.4,P<0.01).B95-8 like subtype was the dominant type in TW.Conclusion High frequency of E2 A in Epstein-Barr virus positive nasopharyngeal carcinoma suggests that E2 A may be associated with the occurrence of the cancer.B95-8-like subtype is the dominant type in healthy people.EBNA2 gene mutation has a tumor correlation rather than a regional correlation.
