目的 探讨表没食子儿茶素没食子酸酯(EGCG)对慢性情绪应激焦虑小鼠海马内γ-氨基丁酸A(GABAA)受体表达的影响.方法 雄性昆明种小鼠60只,随机分为正常对照组、模型组、地西泮组和EGCG高、中、低剂量组,每组10只.正常对照组不接受任何处理,自由饮水和摄食;模型组给予等容积生理盐水;地西泮组给予地西泮5 mg/(kg·d);EGCG高、中、低剂量组分别给予EGCG 60、30、15 mg/(kg·d),各组均灌胃给药.给药第4周末,应用高架十字迷宫检测小鼠焦虑样行为,免疫组织化学方法测定小鼠海马CA1、CA3区GABAA受体的表达.结果 与模型组相比,正常对照组、地西泮组和EGCG中剂量组进入开放臂次数比例(OE%)、开放臂停留时间比例(OT%)均显著增高(F=2.02、5.26,q =3.23~4.60,P<0.05),小鼠海马CA1、CA3区GABAA受体阳性细胞数量显著增加(F=2.86、5.13,q=3.22~4.63,P<0.05);EGCG低剂量和高剂量组OE%、OT%以及海马内GABAA受体阳性细胞数量有增高趋势,但与模型组比较差异无显著性(P>0.05).结论 EGCG具有显著抗焦虑作用,以中剂量作用最为明显,推测EGCG可能是通过增加GABAA受体表达发挥抗焦虑作用.
EFFECT OF EPIGALLOCATECHIN GALLATE ON THE EXPRESSION OF GABAA RECEPTOR IN HIPPOCAMPUS OF CHRONIC EMOTIONAL STRESS-INDUCED MICE
Objective To study the effects of epigallocatechin gallate (EGCG) on the expression of GABAA receptor in hippocampus of chronic emotional stress-induced mice.Methods Sixty male SPF Kunming mice were evenly randomized to normal control group,model group,Diazepam group,high,moderate and low-EGCG-dose groups.The mice in normal control group were not specially treated,drinking and feeding freely as usual;those in the model group were given isometric normal saline;those in Diazepam group were given Diazepam 5 mg/(kg · d),and those in the high,moderate-and low-EGCG dose groups were given 60,30 and 15 mg/(kg · d) of EGCG,respectively.An intragastric administration was conducted in rats of each group.On the end of the forth week,the anxious behavior of the mice was tested adopting Elevated Plus-maze.Employing immunohistochemistry,the expression of GABAA receptor in CA1 and CA3 region of hippocampus of mice was detected.Results Compared with the model group,the time spent in the open arms (OT%),the percentage of open arms entries (OE%) in normal control group,diazepam group and moderate EGCG dose group were significantly increased (F=2.02,5.26;q=3.23 4.60;P<0.05).The number of GABAA receptor positive cells in CA1 and CA3 of hippocampal area increased remarkably (F=2.86,5.13;q =3.22-4.63;P <0.05);the OE%,OT% and GABAA receptor positive cells in hippocampi of low-and high-EGCG dose groups showed an increased trend,but the difference was not significant as compared with the model group (P>0.05).Conclusion Epigallocatechin gallate has a significant anti anxiety effect,the most obvious effect is in its middle dose,presumably,it plays a role in anti anxiety may be by increasing the expression of GABAA.
