EXPRESSION OF ATPASE FAMILY AAA DOMAIN-CONTAINING PROTEIN 2 IN TISSUES OF DIFFERENT LIVER DISEASES AND ITS CLINICAL SIGNIFICANCE
Objective To investigate the expression of ATPase family AAA domain-containing protein 2 (ATAD2) in the tissues of different liver diseases and its clinical significance.Methods A total of 180 samples of different liver lesions obtained by surgical resection and stored in Department of Pathology in The Affiliated Hospital of Qingdao University from April 2,2011 to May 29,2016 were collected,and immunohistochemistry was used to measure the expression of ATAD2.The expression of ATAD2 was compared between the tissues of different liver diseases,and the association between ATAD2 expression in hepatocellular carcinoma and clinicopathological features was analyzed.Results Of all 60 surgical specimens of hepatocellular carcinoma,56 (93.33%) had a mean ATAD2 expression level of (24.87+±22.21) %.Of all 20 surgical specimens of cholangiocellular carcinoma,17 (85.00 %) had a mean ATAD2 expression level of (21.45 ± 19.69)%.Four specimens of high-grade hepatic dysplastic nodules had a mean ATAD2 expression level of (1.00±0.70) %,and no ATAD2 expression was observed in low-grade hepatic dysplastic nodules,hepatic adenoma,focal nodular hyperplasia,liver cirrhosis,and normal liver tissue.The hepatocellular carcinoma tissue had a significantly higher mean ATAD2 expression level than hepatic dysplastic nodules,hepatic adenoma,focal nodular hyperplasia,liver cirrhosis,and normal liver tissue (F=24.45,q=7.37-13.36,P<0.01),while there were no significant differences in ATAD2 expression rate and mean ATAD2 expression level between hepatocellular carcinoma tissue and cholangiocellular carcinoma tissue (P>0.05).Among the 56 surgical specimens of hepatocellular carcinoma with ATAD2 expression,52 (92.86%) had a mean expression level of ≥3%,and 50 (89.29%) had a mean expression level of ≥5%.Among the 38 surgical specimens of grade I-Ⅱ hepatocellular carcinoma,35 (92.11%) had a mean expression level of ≥1%,31 (81.58%) had a mean expression level of ≥3%,and 29 (76.32%) had a mean expression level of ≥5%.ATAD2 expression level in hepatocellular carcinoma was associated with serum alpha-fetoprotein level,presence or absence of hepatitis B surface antigens,and presence or absence of liver cirrhosis (t =2.09-2.30,P<0.05).Conclusion ATAD2 is highly expressed in hepatocellular carcinoma tissue and may play an important role in the development of hepatocellular carcinoma.It may also be involved in malignant transformation of cells.Measurement of ATAD2 expression has a high reference value in evaluating the nature of liver lesions in clinical practice.
