ANALYSIS OF SPECIFIC PROTEINS IN NERVE REGENERATION CONDITIONED FLUID AT DIFFERENT CRITICAL TIME POINTS
Objective To investigate the specific proteins in the nerve regeneration conditioned fluid (NRCF) at four critical time points and its relationship with the peripheral nerve regeneration in different stages of nerve injury repair.Methods A peripheral nerve regeneration chamber (NRC) model was established and the NRCF samples were collected at 3,7,14,and 25 d after transection of the sciatic nerve.The NRCF samples were analyzed using SDS-PAGE electrophoresis,isobaric tags for relative and absolute quantitation,and mass spectrometry.Results The protein bands of NRCF samples at the four time points were uniform and present with good parallelism.A total of 876 proteins were identified and the numbers of specific proteins were 95,137,18,and 58,respectively,at 3,7,14,and 25 d after transection of the sciatic nerve.Conclusion There are rich and many types of proteins in the NRCF.The specific proteins at each critical time point are closely related to peripheral nerve regeneration.
