摘要
目的:在耐药胃癌细胞系SGC7901/ADR中对Ro 60进行克隆,对Ro 60在耐药胃癌细胞系SGC7901/ADR和药敏胃癌细胞系SGC7901中的表达情况进行比较分析.方法:应用RT-PCR法克隆Ro 60的编码基因,通过DNA序列测定对所克隆的基因进行验证.应用半定量RT-PCR法检测Ro 60在耐药胃癌细胞系SGC7901/ADR和药敏胃癌细胞系SGC790l中的表达情况.结果:在耐药胃癌细胞系SGC7901/ADR中,成功克隆了Ro 60的编码基因,其DNA序列与Ro 60的cDNA序列完全一致.半定量RT-PCR结果显示,Ro 60在耐药胃癌细胞系SGC7901/ADR中的表达强度显著高于药敏胃癌细胞系SGC7901细胞中的表达强度,P<0.01.结论:R0 60在耐药胃癌细胞系中高表达,该分子可能是一种胃癌多药耐药相关分子.
Abstract
OBJECTIVE:To Clone Ro 60 encoding gene in drug resistant gastric cancer cell line SGC7901/ADR, and detect the expression levels of Ro 60 in SGC7901/ADR. METHODS: Ro 60 encoding gene was cloned by RT-PCR method t and validated by DNA sequencing. Using semi-quantitative RT-PCR method, the expression levels of Ro 60 in drug resistant gastric cancer cell line SGC7901/ADR and drug sensitive gastric cancer cell line SGC7901 were analyzed. RESULTS: Full length of Ro 60 was cloned in SGC7901/ADR, its sequence was identical with the cDNA of Ro 60. The results of semi-quantitative RT-PCR show that the expression level of Ro 60 in SGC7901/ADR was higher than that in SGC7901 (P<0. 01). CONCLUSION: Ro 60 was highly expressed in drug resistant cancer cell line, and it may be a kind of multi-drug resistance related molecule.
